"The right to search the truth implies also a duty; one must not conceal any part of what one has recognizedto be true."Albert EinsteinFOREWORDThe aims for this book are twofold: (1) to provide the reader with sufficient background material for the understanding of the role that gluconeogenesis plays in health and in diabetes, and to ensure familiarity with the major hormones which regulate the pathway and the methodologies commonly employed for studying it; (2) to supply information on current findings, approaches, and unresolved problems related to the acute regulation of hepatic gluconeogenesis.In selecting the material for the latter part three guidelines were applied:1.The emphasis was on the physiologically important hormones. Thus, the effects of glucagon are presented in more detail than that of other stimulatory hormones. Some hormones known to affect the rate of gluconeogenesis such as growth hormone, thyroid hormone, vasoactive intestinal peptide (VIP), etc., are not discussed because their primary physiological functions are other than the stimulation of gluconeogenesis. The peripheral effects of glucocorticoids, namely the mobilization of gluconeogenic substrates though crucial for gluconeogenesis are not discussed.2.In presenting the material on the hormonal effects on the gluconeogenic pathway, a wide representation of the material was sought. In trying to include as many authors as possible, some duplication of the material was inevitable. This shortcoming seemed preferable over the presentation of the material from only one point of view. Even so, important contributions to the subject might not be represented. I apologize for that.3.Hormonal effects on the liver (the relevance of which for gluconeogenesis has not yet been proven) are presented. This was done in order to be inclusive and unbiased in the selection of the material. There is sometimes a tendency to emphasize observations which lend themselves to easy incorporation into unambiguous, clear-cut models or working hypotheses. An effort was made not to do so.Lastly, 1 would like to express my gratitude to all the contributors to the book for their willingness to spend time and effort in participating in this project.Naomi Kraus-Friedmann
